Adherent culture process for lentivirus——Fixed-bed reactor Culturing lentivirus in a fixed-bed reactor involves several steps to ensure high yield and quality of viral vectors. Fixed-bed reactors are advantageous for this purpose due to their ability to provide a controlled environment and efficient mass transfer. OBiO Tech has equipped 200 ~ 600 m2 fixed-bed reactors in its Lingang facility, provides an adherent culture scale-up solution. Preparation of the Fixed-Bed Reactor Reactor Selection-Sterilization-Seeding Matrix. 2. Cell Preparation Cell Line Selection: Choose a suitable producer cell line for lentivirus production, commonly HEK293T cells. Cell Expansion: Expand the cell line in T-flasks or roller bottles to obtain a sufficient number of cells for seeding the reactor. 3. Seeding the Reactor Cell Inoculation: Inoculate the cells into the fixed-bed reactor. Ensure even distribution of cells within the bed to maximize cell growth and virus production. Culture Medium: Use an optimized culture medium to support cell growth and virus production. This often includes DMEM supplemented with FBS, antibiotics, and other necessary nutrients. 4. Culturing Initial Cell Growth Phase: Allow the cells to attach and grow on the fixed bed. This phase may take several days. Infection Phase: Transduce the cells with the necessary plasmids for lentivirus production. Typically, this includes plasmids encoding the lentiviral vector, packaging plasmids, and envelope plasmids. Culture Conditions: Maintain optimal culture conditions, such as temperature (37°C), CO₂ concentration (5%), and pH (7.2-7.4). Medium Exchange: Periodically exchange the medium to provide fresh nutrients and remove waste products. 5. Virus Harvesting Collection: usually 48-72 hours post-transfection,. Harvest Strategy: Use a perfusion system to continuously or intermittently collect the viral supernatant while fresh medium is supplied to the reactor. Clarification: Filter the collected supernatant to remove cell debris and large particles. 6. Downstream Processing Concentration-Purification-Sterilization 7. Quality Control Titer Determination: Measure the viral titer using methods like qPCR, flow cytometry, or functional assays. Sterility Testing: Ensure the preparation is sterile by performing bacterial and fungal contamination tests.
