New research helps uncover how chemokines cause fast rolling lymphocytes in the vascular system to arrest – an important aspect of immune surveillance. The authors introduce biosensors for extracellular calcium and integrin activation and apply them to demonstrate that chemokine CCL25-triggered calcium influx reduces extracellular calcium levels to the point of allowing fast integrin activation and arrest. #LiveCell #Confocal #Microscopy for this study was performed using a Nikon A1 confocal system. Genetically encoded calcium indicators #GECIs are important for #LiveCell #Microscopy of calcium dynamics, but existing options generally aren’t well suited to detecting low concentrations. Researchers now introduce ultrahigh-affinity GECIs, exploiting a novel strategy for increasing affinity via linker length optimization in GECIs based on circularly permuted fluorescent proteins.
